I take it you're referring to the Forhan Locking wrap? http://www.rodbuilding.org/library/forhan.html
Give it a shot. Won't hurt anything.

The Forhan Locking Wrap is the best locking wrap I've seen so far. I use it for all single foot guide wraps from the smallest, 4mm. B/CLAG Alconite, to the largest, a 30mm. spinning guide. When I wrap a single foot ceramic fly guide as a tip guide the FLW is necessary, IMO, to hold the tip in place as strongly as possible.

As RS1 stated, it certainly won't hurt, but it's also not necessary for a fly rod. This is one of those "personal preference" issues where everyone's answer is correct. Take a look at some factory rods with single foot guides and see how many have locking wraps.

If it gives you a sense of security, then go for it. Personally, I don't use them, and nothing bad has happened. If your guides are wrapped with the proper tension, and your finish extends a bit onto the blank, the guide is not going to move unless it is damaged somehow. I hammered my 1wt all summer and fall, and the SFG's are just fine...no locking wrap.

There is a DVD on building rods that will help you in everything step you need to know.
I bought this DVD and I am very happy with it. Hope this helps.

The Forhan, or other, locking wrap is not necessary if you use old fashioned snake guides or any double footed guide, But is necessary for any single foot guide. Not personal preference, just common sense. It will take a bit More effort to damage a Forhan Locking Wrap protected guide than an unprotected (normal) wrap. Joe, I take good care of my equipment too, but I sell a few rods and fewer of them come back for repairs with the Forhan Locking Wrap than normally wrapped single foot guides. About seven to one by my records. This may be a 'subjective' subject if you are building for yourself but I warrant my work and the less time I spend on free repairs the better I like it.
